TRANSPARENCY OF ACCESS IN PEER-TO-PEER FILESHARING NETWORKSWith the use of peer-to-peer file sharing networks like BitTorrent or Freenet, the distribution of files amongst participants has become fast and easy. In a collaborative environment, this kind of data pool often makes sense, but with the drawback that someone who has provided data to the pool, loses control about who had accessed what file. This transparency of access is often required, e.g. for billing purposes or to fulfil the transparency rule of the new general data protection regulation. There are several options to solve this problem including the use of central systems or logging that involves the source of data. Usually these options introduce single points of failure. The Luxembourg Institute of Science and Technology offers a blockchain based method that is filed as a patent (patent pending) that avoids these single points of failure but still offers the benefits of redundant distributed file storage and still being able to audit selective the access of files.

Transparent wood-based material with high tensile strengthBio-inspired materials are gaining more and more interest, especially as alternatives to plastic. Biominerals such as seashells and pearls are materials generated by living organisms and composed of both organic and inorganic matter which gives them high toughness. However, such materials are usually not transparent, a highly desired property when it comes to the materials we use for various purposes. In general, it is difficult to achieve both high strength and optical transparency in a single composite material. Usually silica-based transparent glass is used as one of the most dominant engineering materials. However, even advanced oxide glasses are highly brittle and fail to resist the fracture. Wood, on the other hand, is crack-resistant and lightweight, but is not transparent. Attempts to develop transparent wood have been made, but failed to meet the requirements set forth by engineering applications. Therefore, professor Kamitakahara’s team developed a new method of producing a wood-based biomineral-inspired material that has high specific strength and toughness as well as transparency.

While generally highly effective, they are associated with adverse effects, and risk of tolerance, dependence, and abuse. The group of Prof. Mike Fainzilber characterized the role of Importina3 and its downstream AP1 mediators in chronic pain. Knockout or knockdown of Importina3 and other components along the pathway was associated with decreased sensitivity to pain cues. Furthermore, inhibition of the Importina3 pathway with specific inhibitors or repurposed drugs provided relief from acute and chronic pain. These findings present novel non-opioid therapeutic targets with potential modalities for intervention.

Treatment for Acute liver failure (ALF)Acute liver failure (ALF) is a rapid deterioration of liver function, resulting in a high mortality rate in the absence of liver transplantation. ALF treatment remains highly limited and mostly supportive due to a lack of a comprehensive understanding of the molecular base of ALF. The group of Profs. Eran Elinav and Ido Amit used high-resolution single-cell analysis methods to unravel the entire molecular mechanism that drives ALF progression. They show that ALF results from activation of the MYC signaling pathway in specific hepatic cells in response to gut microbiome signals. Based on their findings, inhibition of MYC, TLR, or MAPK pathways or microbiome modulation by antibiotics could attenuate ALF.

Thus, there is a clear need for novel therapeutic strategies to overcome resistance, especially in the EGFR+ cohort of patients. The group of Prof. Yosef Yarden has developed a new therapeutic antibody that blocks a common drug resistance mechanism. This antibody completely inhibited tumor progression and was proven especially efficient when administered in combination with known drugs by inhibiting drug resistance thus preventing relapses of NSCLC.

Treatment for Inflammatory Diseases and CancerThe development of kinase inhibitors to treat inflammatory diseases was proved beneficial, however, designing specific inhibitors with minimal side effects and prolonged efficacy is challenging as drug resistance often develops. The current technology includes inhibitors that specifically target the nuclear translocation of p38 kinase, thus inhibiting only its pro? inflammatory activities without developing drug resistance. Two types of inhibitors were developed – a uniquely designed peptide and small molecules. The inhibitors were tested in mice models and proved to effectively treat inflammatory bowel disease and prevent chronic inflammation-driven tumor development.

The present technology identifies a new regulator protein that is essential for the survival of CLL cells.
