Intestine-on-a-chip for Drug Metabolism and Infectious Disease ModelingMicrofluidic organ-on-a-chip technique has recently gained attention for modeling various organs, including the small intestine. A variety of intestine chip designs have been implemented with the double-layer channel separated by a porous PDMS membrane as the most popular option. Available on the market devices are usually composed of epithelial cells in the top channel, endothelial cells at the bottom, and mediums flowing through the two channels to recreate shear stress the intestinal cells experience postnatally. The fluid flowing through the bottom channel is normally designed to emulate the blood. However, the researchers at Kyoto University suspected that mimicking interstitial flow, essential for embryonic organogenesis, in the bottom channel instead could be the key to producing a physiologically relevant intestine-on-a-chip model. They tested this hypothesis and, using human pluripotent stem cells (PSCs), successfully developed a micro-small intestine system that faithfully recapitulates small intestine physiology.

It not only unlocks a new way of reading through listening, but also provides further assistance through audio navigation and visual highlighting of the synchronised text and audio. The product is called iSinkwe, which is the isiZulu name for a bush baby, an animal known for its big eyes and ears. This symbolises how the software empowers the eyes and ears of aspiring readers. Isinkwe is packaged as three subproducts that revolve around the EPUB 3 publishing standard: the converter, augmenter and reader. The converter converts ebooks in other file formats, such as DOCX and PDF, to EPUB 3. The augmenter uses the CSIR’s Qfrency text-to-speech technology to synchronise the text and audio of EPUB 3 ebooks in any of the 11 official South African languages. The audio can be human-narrated by a voice artist or computer-synthesised by Qfrency. The reader reads the synchronised EPUB 3 ebooks, with options to play back and navigate through the audio, as well as highlight the text as the audio is played. “We are targeting the local market initially, since our human language technology expertise currently lies in the context of the South African languages. iSuRe-Cre as a new genetic tool to modify and understand gene functionMost analysis of gene function in biomedical research relies on the use of Cre-lox technology. Since its introduction in 1994, this technology has revolutionized biomedical science because it allows scientists to eliminate or activate the function of any gene in any cell type in the mouse. Because the Cre-loxP recombination event is invisible, genetic and fluorescent reporters have been developed to track Cre activity, so that cells reaching a certain threshold of Cre activity are labeled. However, the main problem is that often the Cre activity level is insufficient to fully recombine and eliminate expression of the target gene, generating uncertainty about whether the desired genetic modification has been achieved. CNIC researchers have developed a method that increases the efficiency and reliability of the analysis of Cre-dependent gene function. Moreover, the use of this genetic tool permits the induction of multiple genetic deletions in the same cell, to allow the study of functional genetic interactions.
