This invention provides a novel method for transplanting target cells into a subject by creating a sealed transplantation space using an acellular membrane attached to a visceral organ. This technique allows for the insertion of a mixture of support cells and target cells, enhancing cell survival and integration. For example, this method can be applied to improve the engraftment of insulin-producing beta islet cells for diabetes treatment, potentially increasing the success rate of cell transplantation.
BACKGROUND
The field of cell transplantation faces significant challenges, particularly in preventing the loss of transplanted cells due to immune reactions and inadequate microenvironments. Current methods, such as portal vein infusion for islet transplantation, often result in high cell loss due to instant blood-mediated inflammatory reactions (IBMIR). This invention addresses these challenges by providing a new approach that minimizes exposure to blood and inflammatory responses, thereby improving the efficiency of cell engraftment.
