The 96-Puddle Paper Plate (96-PPP) is a lab-on-paper analytical platform designed to resolve spatially heterogeneous reaction behaviors that are obscured in conventional bulk well-plate assays. Traditional liquid-filled wells collapse diffusion, convection, and localized reaction dynamics into a single averaged signal, often resulting in poor reproducibility, misleading interpretation, and limited quantitative reliability.
The proposed platform uses silk-screened PDMS barriers on a cellulose substrate to form uniform, leakage-free micro-puddles arranged in a standard 96-well format. This structure preserves spatial reaction significance while remaining fully compatible with existing microplate readers, fluorescence microscopes, and colorimetric imaging systems. Quantitative agreement with conventional 96-well plates has been experimentally validated, while achieving significantly lower signal variation across individual puddles.
By enabling structured reaction confinement, the platform supports time-resolved observation of condition-dependent reaction dynamics, including enzymatic reactions, aggregation processes, redox reactions, and metabolite sensing. Its paper-based, disposable, and storage-efficient design allows low-cost manufacturing, easy scalability, and application-specific customization.
