This invention provides a sensitive and specific LC-MS-MRM method for quantifying outer-arm and core fucosylated N-glycopeptides directly in plasma or serum without sample fractionation or enrichment. It utilizes optimized low-energy fragmentation to produce linkage-specific Y-ions as MRM transitions, enabling quantification of fucosylation types across multiple glycoforms of plasma proteins. The method demonstrates increased outer-arm fucosylation in liver cirrhosis patients compared to healthy controls.

BACKGROUND

Altered fucosylation of N-glycoproteins is associated with liver diseases, but existing methods struggle to quantify core vs. outer-arm fucosylation site-specifically. This invention overcomes technical challenges to achieve sensitive, linkage-specific quantification of fucosylated glycoforms directly in complex samples.
