Description of the technology offer :

The UCA research group on “Biological and Enzymatic Reactors” (TEP-105) has developed a novel culture medium formulated specifically for the bioproduction of ethanol and hydrogen from glycerine, through the bacterial activity of the species Escherichia coli. It is a simple and attractive method in which the source of carbon employed is glycerine, obtained as a by-product in the biodiesel industry, that is available in large quantities and hence at low price.

Exploitation of the large amounts of glycerine produced as a by-product of the production of biodiesel and similar industrial processes.
The employment of this culture medium increases the production yield of ethanol and hydrogen, in comparison with that obtained from the use of other sources of carbon.
The effect of optimizing the culture medium is to reduce the costs and save energy in the industrial-scale fermentation process.
No other alternative culture media exist that have been formulated and optimized for E. coli that would allow greater production yields of ethanol to be obtained.
It represents a simple and attractive way of producing hydrogen and ethanol from a carbon source of low price and in plentiful supply.
The high degree of reduction of the glycerine, in comparison with sugars, allows reduced chemical compounds such as succinate, xylitol, propionate, hydrogen and others to be obtained. Read more

The proposed technology consists of a new microorganism with demonstrated biopreservative activity. Its use in the fruit industry, particularly in minimally processed fruit, prevents the development of food-borne pathogens, ensures food safety and maintains the product quality by means of an easy-friendly method. Collaborations with industrial partners in order to fulfill the development of a commercial product are sought.

This new technology consists of a new microorganism with demonstrated biopreservative activity, capable of inhibiting and even eliminating the growth of food-borne human pathogens such as Salmonella, Listeria monocytogenes and Escherichia coli O157: H7 in fresh-cut fruit. Its effectiveness has been demonstrated within 5 – 20ºC. It does not grow at 37ºC, it does not survive in gastric juice and there are no references about potential harmful effects in humans.

Nowadays, the consumption of fresh-cut fruit is increasing throughout Europe and USA. Although these products undergo minimal processing, the risk of contamination by human pathogenic bacteria may be increased during industrial and commercial handling.

Disinfection by means of chemical methods and/or good manufacturing practices does not assure the complete elimination.The use of the proposed technology prevents the development of food-borne pathogens, ensures food safety and maintains the product quality throughout the product shelf-life by means of an easy-friendly method. This work is performed using DNA provided by the client. We use prokaryotic expression systems (Escherichia coli and Bacillus subtilis) as well of a yeast expression system (Pichia pastoris). We apply high standards in implementing quality services both technically and procedurally, that ensures high purity of the proteins we produce.

In Zurko Research we perform cloning, expression and purification of proteins in bacterial or yeast systems. We develop new enzymes with interesting activities for the industry and compare them with commercial enzymes.

In Zurko Research we develop pilot tests of these new enzymatic activities not yet described for its use in the industry. We also search for new enzymes with industrial interest focusing in improving bioprocesses and biotransformations. We optimize different complex proteolytic bioprocesses for better utilization of by-products following European standards in quality and in environment impact. We investigate the valorisation of by-products and residues, searching for the economic convenience of the company first. We conduct pilot tests to obtain improved formulations of active peptides and amino acids with potential applications in different industrial sectors: feed, biomedicine, plant, food, etc.

APPLICATIONS IN THE DAIRY INDUSTRY

1) Ripening of cheeses: Lipases are used to accelerate the ripening of blue cheeses. For the production milk rennet, renins and pepsins are used.

2) The use of enzymes in pastry and bakery such as Amylases, beta-Glucanases, Hemicellulases, and Xylanases produce more sugars for the fermentation and improve the quality and taste of the bread and bakery products.

3) Total Hydrolyzed Whey for functional foods. By total biohydrolysis of whey an hydrolysate is obtained with high content of essential amino acids suited for human nutrition. Exo and broad spectrum Endoproteasas are used.

4) For a clarifying effect of fruit juices and vegetables using various enzymes: Hemicellulases, Pentosanases, Cellulases, Xylanases and Pectinases. To remove the bitter taste of grapefruit juice because of the citrus peel alpha-L-Rahmanosidase and beta-Glucosidase are used.

APPLICATIONS IN THE PRODUCTION OF FEED

Production of feed products highly digestible. This can be achieved with the addition of certain enzymes in the animal feed, such as Glucanases, Xylanases, Amylases and Phytases (better absorption of phosphate)

1 .- Use of enzymes for the degradation/treatment of cererals. Among these enzymes are: Amylases, Xylanases, Gluconases and Cellulases, mainly from microbial source.

2 .- Use of microbial proteases to hydrolyze proteins, to obtain highly valued amino acids and peptides.

3 .- Degradation of phytate by using Phytase of bacterial origin.

APPLICATIONS IN MEAT AND FISH INDUSTRIES

1) For tender and tasty meats. By enzymatic treatment of certain meat cuts using papain, Endo and Exo Peptidases as well as Acid Proteases a more tender meat is obtained.

2) total hydrolysate from meat for functional foods. By total biohydrolysis of meat and fish, an hydrolysate is obtained with high content of essential amino acids for human nutrition. Exo and broad spectrum Endoproteasas are used, as well as Collagenases.

3) What to do with category 3 meat waste?
The use of enzymes on of hard to degrade waste and residues (hooves, horns, hair, fur, etc.) from meat processing plants, produces free amino acids and bioactive peptides with important applications in phytosanitary. We use enzymes such as Collagenases, Pz-Peptidases, Keratinases and Subtilisin.

A group of researchers from a Portuguese university has developed a process to obtain porcelain stoneware tiles with anti-bacterial activity, namely, that promotes the bacteria’s death while all the other required technological characteristics, particularly the chemical resistance and anti-staining properties, are assured. In comparison with the existing solutions the two advantages of this process are: (i) the use of a common raw material in the porcelain stoneware tiles processing and (ii) the no need of radiation being only necessary the direct contact with the bacteria. The developed material can be used in the revetment of places where the anti-bacterial properties are relevant (hospitals, kindergartens, etc.).

This strain is capable to reduce the risks and improve the health and quality of life of individuals with coeliac disease and other disorders related to gluten ingestion. The strain is capable of taking up and hydrolysing the gluten peptides responsible for the disorders. This strain has immunomodulatory properties. The strain is also capable of inhibiting pathogenic bacteria.

Gluten proteins (gliadins and analogous prolamins and glutenins) represent the main environmental factor that triggers coeliac disease and other related disorders. These proteins contain peptide sequences rich in proline and glutamine, making them more resistant to digestive enzymes than other proteins in the diet and,therefore, capable of remaining in the intestinal lumen. In genetically predisposed individuals, these peptides are responsible for an anomalous reaction that involves both the innate and the adaptive immune system and that, globally, lead to chronic inflammation of the intestinal mucosa, increased intraepithelial lymphocytes, crypt hyperplasia, and a progressive deterioration of intestinal villi including their total disappearance.

The composition of the intestinal microbiota of celiac disease patients also presents an imbalance in relation to that of healthy control individuals, characterised by the predominance of proinflammatory bacteria and a reduced proportion and composition of lactic acid bacteria and bifidobacteria.

A new strain of the Bifidobacterium longum (CECT 7347), its cellular components, secreted molecules and compounds resulting from its metabolism and any combinations thereof with each other and/or with other microorganisms or bioactive compounds, in the form of preparations were designed to reduce the risks and improve the health and quality of life of individuals with coeliac disease. For example fission yeast can be cultivated in a low pH environment up to pH 3 differentiating it from other organisms. If a protein and/or reaction require such conditions fission yeast is a very well suited model organism.

At comparatively low cost the PomBioTech researchers will make a proof-of-concept study to check whether your protein of interest can be functionally produced by recombinant S. pombe.
